To set up Microsoft Expression Web 2 to automatically open your last edited website, you need to change the startup options for Expression Web.
Go to Tools > Application Options… to open the Application Options dialog box as shown below:

On the General tab, under the Startup section, check the preference ‘Open last Web site automatically when Expression Web starts’ and click on OK. Turn this option off if you do not want Expression Web to open the last website you were working on when Expression Web is launched.

Checking that box doesn’t load EW2 at startup for me. Not sure why.

The article ‘Automatically open last edited website in Microsoft Expression Web 2′ – helps you set up Microsoft Expression Web 2 to automatically open your last edited website, when EW2 is launched.
To automatically load EW2 at startup (of your system) you will have to put a shortcut to EW2 in this folder –
c:\users\username\appdata\roaming\microsoft\windows\start menu\programs
